چکیده

Interleukin (IL)-6 is an autocrine growth factor for renal cell carcinoma (RCC). We sought to determine whether p53 regulates constitutive IL-6 production. RCC cell lines containing mutant (mut) p53 produced higher levels of IL-6 than those containing wild-type (wt) p53 (P < 0.05). Transfection of wt p53 into RCC cell lines bearing mut p53 (UOK 121LN) or wt p53 (A498 and ACHN) resulted in repression of IL-6 promoter chloramphenicol acetyltransferase activity (P < 0.05). Mutant p53 was either less effective at repressing IL-6 promoter activity (ACHN cells) or enhanced IL-6 promoter activity (A498 cells). A498 cells stably transfected with mut p53 produced higher levels of IL-6 than A498 cells transfected with an empty expression vector (P < 0.05). Electrophoretic mobility shift assays showed decreased binding of CAAT enhancer binding protein, cyclic AMP responsive element binding protein, nuclear factorB transcription factors to the IL-6 promoter in various RCC cell lines transfected with wt p53 (P < 0.05) but not in those transfected with mut p53. These data suggest that: (a) mutation of p53 contributes to the overexpression of IL-6 in RCC; and (b) wt p53 represses IL-6 expression, at least in part, by interfering with specific transcription factor binding to the IL-6 promoter.

Dyspnea as the First Manifestation of Silent Renal Cell Carcinoma

Renal cell carcinoma (RCC) accounts for only 3% of adult malignancies, and the lung is the most common site of metastasis of this tumor, which may be accompanied by pleural metastasis. However, solitary pleural involvement is very rare and its presentation with dyspnea as the first manifestation of RCC is extremely rare.We describe a 39-year-old male with episodes of dyspnea dating back 6...

Expression Of P53 And Ki67 Proteins In Renal Cell Carcinoma And Its Relationship With Nuclear Grade

Background and Objective: Evaluation of tumor proliferative activity may provide a predicting parameter to estimate biologic aggression and a subsequent prognosis that has been evaluated in many malignancies. We have selected renal cell carcinoma (RCC) in this study. To determine tumor proliferative activity, KI67 antibody was applied and results were compared with apoptosis, applying P53 antib...

Prognostic value of p53 in renal cell carcinoma

Background and Objectives: RCC is one of the most common genitourinary cancers. Accurate prediction of prognosis would be valuable for adjuvant trial design, counseling and effectively scheduling follow up visits. P53 is a tumor suppressor gene that expresses a protein that involved in both cell-cycle ar...

Feasibility of sentinel lymph node mapping in renal cell carcinoma using intraoperative radiotracer injection

Introduction: The applicability of sentinel lymph node biopsy for early detection of metastasis in patients with renal cell carcinoma (RCC) is still in the validation phase and under investigation, which might be due to the unpredictability of the lymphatic pattern in RCC. In this study, we aimed to evaluate the feasibility and accuracy of sentinel node biopsy in patients with ...
